Abstract

DETERMINATION OF ANTIOXIDANT CONTENT OF FEW AROMATIC PLANTS BY RP-HPLC METHOD

ABSTRACT

A rapid, simple and precise high-performance liquid chromatographic (HPLC) method coupled with photo diode array detector was developed for the simultaneous determination of gallic acid, catechin and caffeic acid in aromatic tulsi plant. The chromatographic method used Micrbondapak C18 column as the stationary phase, 0.2% ortho phosphoric acid in water and acetonitrile at a ratio (81:19) as the mobile phase. The flow rate was maintained at 1.2 ml/min and wavelength of detection was set at 225 nm. The calibration curve showed good linearity with regression coefficient of 0.998, -0.998-0.997 respectively for gallic acid, catechin and caffeic acid in the linear concentration range of 1.6 ?g/ml to 25.6?g/ml. The relative standard deviation of intra-day and inter-day precision for three the analytes was found to be less than 3% and a very good separation of the components was obtained with retention time of 3.22, 4.92 and 6.89 respectively. The d was successfully appliegd to the estimation of caffeic acid, catechin and gallic acid inmethanolic extracts of different species of tulsi plant (Ocimum sanctum and Ocimumtenuiflorim) using the newly developed HPLC method.
